We highlight a recent study exploring the hand‐off of UV damage to several key nucleotide excision repair (NER) proteins in the cascade: UV‐DDB, XPC and TFIIH. The delicate dance of DNA repair proteins is choreographed by the dynamic hand‐off of DNA damage from one recognition complex to another damage verification protein or set of proteins. These DNA transactions on chromatin are strictly chaperoned by post‐translational modifications (PTM). This new study examines the role that ubiquitylation and subsequent DDB2 degradation has during (...) this process. In total, this study suggests an intricate cellular timer mechanism that under normal conditions DDB2 helps recruit and ubiquitylate XPC, stabilizing XPC at damaged sites. If DDB2 persists at damaged sites too long, it is turned over by auto‐ubiquitylation and removed from DNA by the action of VCP/p97 for degradation in the 26S proteosome. (shrink)

During the process of E. coli nucleotide excision repair, DNA damage recognition and processing are achieved by the action of the uvrA, uvrB, and uvrC gene products. The availability of highly purified proteins has lead to a detailed molecular description of E. coli nucleotide excision repair that serves as a model for similar processes in eukaryotes. An interesting aspect of this repair system is the protein complex's ability to work on a vast array of DNA lesions that differ widely in (...) their chemical composition and molecular architecture. Here we propose a model for damage recognition in which the UvrB protein serves as the component that confers enhanced specificity to a preincision complex. We hypothesize that one major determinant for the formation of a stable preincision complex appears to be the disruption of base stacking interactions by DNA lesions. (shrink)

Generalising what is learned about one stimulus to other but perceptually related stimuli is a basic behavioural phenomenon. We evaluated whether a rule learning mechanism may serve to explain such generalisation. To this end, we assessed whether inference rules communicated through verbal instructions affect generalisation. Expectancy ratings, but not valence ratings, proved sensitive to this manipulation. In addition to revealing a role for inference rules in generalisation, our study has clinical implications as well. More specifically, we argue that targeting inference (...) rules might prove to be an effective strategy to affect the excessive generalisation that is often observed in psychopathology. (shrink)

The aim of this review is to show the fruitfulness of using images of facial expressions as experimental stimuli in order to study how neural systems support biologically relevant learning as it relates to social interactions. Here we consider facial expressions as naturally conditioned stimuli which, when presented in experimental paradigms, evoke activation in amygdala–prefrontal neural circuits that serve to decipher the predictive meaning of the expressions. Facial expressions offer a relatively innocuous strategy with which to investigate these normal variations (...) in affective information processing, as well as the promise of elucidating what role the aberrance of such processing might play in emotional disorders. Philosophers and cognitive scientists address the relationships among the senses and the connections between conscious experiences that form unified wholes. In this volume, cognitive scientists and philosophers examine two closely related aspects of mind and mental functioning: the relationships among the various senses and the links that connect different conscious experiences to form unified wholes. This second of three articles outlining the development and practice of a different approach to neurotheology discusses the construction of a suitable methodology for the project based on the work of J. Wentzel van Huyssteen. It explores the origin and contours of his concept of postfoundational rationality, its potential as a locus for epistemological parity between science and religion and the distinctive and unique transversal space model for interdisciplinary dialogue which he builds on these. It then proposes a further development (...) of the model which has the potential to produce a very different type of additional and original dialogical outcome. While such “transversal” outputs may initially seem counter and strange they not only flow naturally from the models’ own inherent dynamics but also open up the possibility of a distinctively different form of neurotheology. Bennett and Hacker criticize a number of neuroscientists and philosophers for attributing capacities which belong to the human being as a whole, like perceiving or deciding, to a “part” of the human being, viz. the brain. They call this type of mistake the “mereological fallacy”. Interestingly, the authors say that these capacities cannot be ascribed to the mind either. They reject not only materialistic monism but also Cartesian dualism, arguing that many predicates describing human life do not refer to (...) physical or mental properties, nor to the sum of such properties. I agree with this important principle and with the critique of the mereological fallacy which it underpins, but I have two objections to the authors’ view. Firstly, I think that the brain is not literally a part of the human being, as suggested. Secondly, Bennett and Hacker do not offer an account of body and mind which explains in a systematic way how the domain of phenomena which transcends the mental and the physical relates to the mental and the physical. I first argue that Helmuth Plessner’s philosophical anthropology provides the kind of account we need. Then, drawing on Plessner, I present an alternative view of the mereological relationships between brain and human being. My criticism does not undercut Bennett and Hacker’s diagnosis of the mereological fallacy but rather gives it a more solid philosophical–anthropological foundation. (shrink)

Background Preserving persons’ dignity is integral to nursing. More research is needed to explore how a diversity of patients, particularly those that experience illness from a young age, experience dignity. Aim Describe the characteristics of dignity for persons living with serious illness. Research design Using a secondary data set of twenty audio-recorded interviews, a thematic content analysis was conducted to identify characteristics of dignity. The research team employed van Gennip et al.’s, 2013 “Model of Dignity in Illness” (1) to create (...) a codebook, which the authors utilized to independently code twenty narrative interview transcripts. Participants and research context Twenty persons living with serious illness of heart failure and/or dialysis-dependent renal failure who were admitted in an acute care hospital. Ethical considerations This study was approved on August 26, 2019, by the Colorado Multiple Institutional Review Board (COMIRB) IRB Protocol #19-1874. Findings Early-onset participants expressed markedly different dignity concerns than late-onset participants. In the individual domain, early-onset participants felt that their illness was “normal”; they did not experience the “healthy person to patient” transition described by older onset participants. In the relational domain, early-onset participants expressed that their relationships had already integrated their illness while late-onset participants felt that their illness harmed many of their relationships. In the societal domain, early-onset participants described dignity concerns related to how society impacted their ability to financially support themselves during their illness. Discussion Differences in the dignity experience of early-onset and late-onset participants are informed by Erikson’s “Model of Development” and by Aranda and Jones feminist critique of dignity in healthcare. Conclusions Persons with early-onset illness experience dignity differently. Awareness of the importance of work and financial independence to the experience of dignity for seriously ill patients may enhance persons’ dignity experience. (shrink)

The Institute on Religion in an Age of Science has a long history of delivering conferences addressing topics of interest in the field of science and religion. The following papers from the 2013 summer conference on “The Scientific, Spiritual, and Moral Challenges in Solving the World Food Crisis” are, in keeping with the eclectic nature of these conferences, very different in content and approach. Such differences underline the challenges of synergistically combining scientific and religious insights to increase understanding of global (...) problems and their possible solutions. This in turn reflects deeper questions about the purpose and nature of the science/religion dialogue. These papers suggest various ways in which the two perspectives can be combined in the pursuit of building better understandings of food-related issues, as well as highlighting difficulties and limitations which need to be addressed if the fruits of such dialogue are to make a wider impact. As such they serve as useful pointers for how this type of science/religion interaction might be further developed and deployed. (shrink)

Talk of ‘brains deciding’, ‘brains inferring’ or ‘brains perceiving’ is common in contemporary cognitive science and neuroscience. A number of authors (most notably Peter Hacker and Maxwell Bennett) argue that such talk commits a category mistake; deciding, making inferences or perceiving are abilities properly ascribed to humans and not to human subsystems or subparts. Here I will argue that ascribing such properties to human brains or cognitive systems within the brain is proper. I will argue that ‘inferring’ or ‘perceiving’ (...) are best conceptualized as functional terms. Since most functions we know of are multiply realizable, this opens the door for atypical inferences and perceptions that can be ascribed to various organisms and their subsystems. (shrink)

New York and Oxford: Oxford University Press, 1999. Pp. xii + 340. Cloth, $45.00. The author acknowledges his debt to the "great Kant books of the 1960s, Jonathan Bennett's Kant's Analytic, and P. F. Strawson's The Bounds of Sense."Their analytical spirit lives on in this book, but the analyses (...) are fresh, take surprising turns, and make unexpected connections to current concerns. In seventeen chapters Van Cleve deals with necessity and the a priori, space and geometry, incongruent counterparts, time, the ideality of matter, experience and objects, substance in the First Analogy, the Second Analogy, noumena, problems of the self, rational theology, and Kant and contemporary irrealism. To sustain the flow of argument, the discussion of certain details, often important, is postponed and taken up in fifteen appendices, and there are nearly seventy pages of careful notes.Chapter 10, "Noumena and Things in Themselves," (134 ff.) criticizes in detail the prevailing "one world" or "double aspect" view of objects, namely that Kant allows just one set of objects, but two ways of "considering" them. Things in themselves are the very same objects as appearances, but considered as they are independent of our experience of them, a view developed in Henry Allison's Kant's Transcendental Idealism. Against this Van Cleve maintains (and I concur) that "appearances are intentionalia, and that intentionalia are constructions out of states of perceivers" (150). The distinction is between two separate universes of discourse, "not between two ways of discoursing [End Page 448] about the same objects" (ibid.). There is no space to report the sharply argued detail that led him to hold the double aspect view to be unfathomable and incoherent. He takes Kant to be a phenomenalist but holding to a version far removed from the reductionist phenomenalism of Russell, Broad, Price, and others. Here the basic elements are not made out to be indubitable givens, but are, rather, sensations. The structures built on them are, according to Van Cleve, "virtual objects" in the sense in which Quine speaks about "virtual classes." To say that something is an element of a class {F:Fx} is only to say that it is F. So, likewise, to say that, e.g., a patch of red exists is short for saying that a certain representation occurs. Carnap's Kantian masterpiece, TheLogical Structure of the World (The Aufbau) could have been consulted as a comprehensive guide to the kind of virtuality here envisaged.It is Van Cleve's stated aim never to discuss Kantian arguments without making their premise and conclusion structure clear (viii). Quite a few of them he finds persuasive; in other cases the arguments are valid, but one or the other premise doubtful. Sometimes he sides with Kant against his critics, sometimes the opposite, and occasionally, as in the case of the Second Analogy, it remains unclear what Kant's argument was. He has shown in this and earlier publications that all reconstructions are flawed, so that either Kant had no good argument, or his argument has not yet been discovered.The title Problems from Kant must be taken seriously. The book rarely deals with textual problems, historical context, or translation difficulties. Rather, the focus is on perennial and still compelling problems. Themes from the First Critique are tracked to and connected with current concerns and sometimes recently ventilated issues are traced back to Kant, as in the last chapter, where the question is raised if Kant was an internal realist in the style of Putnam, or perhaps an anti-realist like Dummett. The contrast with Kant shows that like all complex positions, these are shopping baskets with several goods in them, some in Kant's inventory, others not.Logical notation is used sparingly, and only where it clarifies—no attempt here to shoehorn arguments into unneeded symbolism. Philosophers and scientists are concerned with the why and the how of things. Questions like the following are so much grist for the philosopher’s and scientist’s mill: How can we be free and yet live in a deterministic universe?, How do neural processes give rise to conscious experience?, Why does conscious experience accompany certain physiological events at all?, How is a three-dimensional perception of depth generated by a pair of two-dimensional retinal images?. Since Belnap and Steel’s pioneering work on the (...) logic of questions, Van Fraassen has managed to apply their approach in constructing an account of the logic of why-questions. Comparatively little, by contrast, has been written on the logic of how-questions despite the apparent centrality of questions such as How is it possible for us to be both free and determined? to philosophical enterprise.1 In what follows I develop a logic for how-questions of various sorts including how-questions of cognitive resolution, how-questions of manner, how-questions of method, of means, and of mechanism. It can be shown by means of a paradox that, given the Principle of Sufficient Reason, there is no conjunction of all contingent truths. The question is, or ought to be, how to interpret that result: _Quid sibi velit?_ A celebrated argument against PSR due to Peter van Inwagen and Jonathan Bennett in effect interprets the result to mean that PSR entails that there are no contingent truths. But reflection on parallels in philosophy of mathematics shows it can equally (...) be interpreted either as a proof that there are "too many" contingent truths to combine in a single conjunction or as a proof that the concept _contingent truth_ is indefinitely extensible and there is no such thing as "all contingent truths." Either interpretation would reconcile PSR with contingent truth, but the natural rationales of those interpretations are at odds. This essay argues that the second interpretation is a more satisfactory explanation of why, if PSR is true, there should be no conjunction of all contingent truths. This sheds new light on the nature of the explanatory demand embedded in PSR and uncovers a number of surprising implications for the commitments of rationalism. (shrink)

PurposeThis paper aims to demonstrate the relevance of worldviews of the global south to debates of artificial intelligence, enhancing the human rights debate on artificial intelligence (AI) and critically reviewing the paper of UNESCO Commission on the Ethics of Scientific Knowledge and Technology (COMEST) that preceded the drafting of the UNESCO guidelines on AI. Different value systems may lead to different choices in programming and application of AI. Programming languages may acerbate existing biases as a people’s worldview is captured in (...) its language. What are the implications for AI when seen from a collective ontology? Ubuntu (I am a person through other persons) starts from collective morals rather than individual ethics.Design/methodology/approachLiterature overview on the African philosophy of Ubuntu as applied to artificial intelligence. Application of it to the United Nations Educational, Scientific and Cultural Organisation (UNESCO) debates on establishing guidelines to the ethics of artificial intelligence.FindingsMetaphysically, Ubuntu and its conception of social personhood (attained during one’s life) largely rejects transhumanism. When confronted with economic choices, Ubuntu favors sharing above competition and thus an anticapitalist logic of equitable distribution of AI benefits, humaneness and nonexploitation. When confronted with issues of privacy, Ubuntu emphasizes transparency to group members, rather than individual privacy, yet it calls for stronger (group privacy) protection. In democratic terms, it promotes consensus decision-making over representative democracy. Certain applications of AI may be more controversial in Africa than in other parts of the world, like care for the elderly, that deserve the utmost respect and attention, and which builds moral personhood. At the same time, AI may be helpful, as care from the home and community is encouraged from an Ubuntu perspective. The report on AI and ethics of the UNESCO World COMEST formulated principles as input, which are analyzed from the African ontological point of view. COMEST departs from “universal” concepts of individual human rights, sustainability and good governance which are not necessarily fully compatible with relatedness, including future and past generations. Next to rules based approaches, which may hamper diversity, bottom-up approaches are needed with intercultural deep learning algorithms.Research limitations/implicationsThere is very few existing literature on AI and Ubuntu. Therefore, this paper is of an explorative nature.Practical implicationsThe ethics of Ubuntu offers unique vantage points in looking at the organization of society and economics today, which are also relevant for development of AI, especially in its tenet of relatedness rather than individuality (and practical use of AI for individuals), taking responsibility for society as a whole (such as analyzing the benefit of AI for all strata of society), and embodying true inclusiveness. Whether looking at top-down guidelines for the development and implementation of AI or the bottom-up ethical learning process of AI (deep learning), ethics of the Global South can have an important role to play to combat global individualist tendencies and inequity, likely reinforced by AI. This warrants far more research.Social implicationsApplications of AI in Africa are not contextualized, do not address the most pressing needs of the African continent, lead to cybersecurity issues and also do not incorporate African ethics. UNESCO’s work in this regard is important but expert inputs are largely centered around Western “universal” principles and Organisation for Economic Cooperation and Development and EU precedence. African ethics have, so far, a small role to play in global ethics and philosophy and therefore risk to be overlooked in the discussion on AI and ethics. This is why the consultation process of UNESCO on ethics of AI was of paramount importance. However, it does not automatically lead to consultation of African philosophers or sages, as many are educated in Western (ized) education systems. See further details under practical implications.Originality/valueThis is a new area of research in which little work has been done so far. This paper offers the opportunity to widen the debate on AI and ethics beyond the conventional discourse, involving multiple worldviews, of which Ubuntu is just one. (shrink)

They provide direct information on how Babylonian scholars conducted scientific research and viewed political, economic, and religious events of their time—in other words, how they experienced their era. The Report's editorial mandate is both to examine issues of current importance and to invite bioethics to broaden the range of issues it probes. Thus along with articles exploring the relationship between patients and doctors, or health policy, or any of the myriad other familiar concerns of ethics in medicine, from time to time the Report has published articles about public health, animal experimentation, or “environmental ethics” broadly construed. The most recent was the special supplement Nature, Polis, Ethics in the (...) November‐December (1998) issue. Some, of course, have argued that “bio‐ethics” ought always to encompass concern for issues beyond the bedside. When the biologist Van Rensselaer Potter proposed the term bioethics nearly thirty years ago, the field he had in mind lay at the intersection of ethics and the biological sciences in general. In light especially of Nature, Polis, Ethics's call for a substantive “humans‐nature ethics” it seems appropriate, therefore, to invite one of the earliest proponents of a generous understanding of bioethics to reflect on the character of the field and the questions with which it should grapple. A total of 80 authors working in a variety of scientific disciplines commented on the theoretical model of CLimate, Aggression, and Self-control in Humans (CLASH). The commentaries cover a wide range of issues, including the logic and assumptions of CLASH, the evidence in support of CLASH, and other possible causes of aggression and violence (e.g., wealth, income inequality, political circumstances, historic circumstances, pathogen stress). Some commentaries also provide data relevant to CLASH. Here we clarify the logic and assumptions of CLASH (...) and discusses its extensions and boundary conditions. We also offer suggestions for future research. Regardless of whether none, some, or all of CLASH is found to be true, we hope it will stimulate future research on the link between climate and human behavior. Problems from Kant is distinctive in the way that it combines the crisp and often extremely critical style of argumentation found in Jonathan Bennett’s work with a very helpful grasp of the much more metaphysical character of the leading trends in current systematic philosophy. Although the book defends a phenomenalist reading of Kant's transcendental idealism that is not far from Bennett's empiricist interpretation, it also stresses many points that derive mostly from a philosophical and interpretative sympathy for the (...) rationalist tradition and a revival of modal considerations in the spirit of David Lewis and Alvin Plantinga. Since there is not enough room here to go into the many points on which I very much agree with the rationalist side of Van Cleve’s reading, I will take this occasion to play the role of critic and concentrate on one of the few areas in which it appears we have some serious disagreements. These disagreements are worthy of special attention because they arise in the chapter that overlaps the least with Van Cleve’s earlier publications, and they concern a topic that all would agree is at the very center of the Critique, namely Kant’s Transcendental Deduction of the categories.
